Metabolic neural mapping in neonatal rats

Abstract: Functional neural mapping by 14C-deoxyglucose autoradiography in adult rats has shown that increases in neural metabolic rate that are coupled to increased neurophysiological activity are more evident in axon terminals and dendrites than neuron cell bodies. Regions containing architectonically well-defined concentrations of terminals and dendrites (neuropil) have high metabolic rates when the neuropil is physiologically active. In neonatal rats, however, we find that regions containing well-defined groupings o… Show more

“…Immediately after conditioning, pups were decapitated and their brains were quickly removed, frozen in 2-methylbutane (Ϫ45°C), and stored in a Ϫ70°C freezer. For analysis, brains were sectioned (20 m) in a Ϫ20°C cryostat, and every other section was saved to be placed on a coverslip and exposed for 5 d along with 14 C standards ( 14 C standards, 10 ϫ 0.02 mCi; American Radiolabeled Chemicals, St. Louis, MO) to x-ray film (DiRocco and Hall, 1981;Coopersmith and Leon, 1986;Sullivan and Wilson, 1995;Nudo and Masterton, 2004).…”

“…Immediately after training, pups were decapitated and their brains quickly removed, frozen in 2-methylbutane (À45°C), and stored in a À70°C freezer. For analysis, brains were sectioned (20 mm) in a À20°C cryostat, with every second section placed on a cover slip and exposed for 5 d along with 14 C standards ( 14 C standards 10 3 0.02 mCi, American Radiolabeled Chemicals, Inc.) to X-ray film (DiRocco and Hall 1981;Sullivan and Wilson 1995). The autoradiographs were analyzed using National Institutes of Health image software that allows pseudocolor imaging and quantitative optical densitometry.…”

“…Brains were removed immediately after conditioning, frozen in 2-methylbutane (‫°54מ‬C) and stored at ‫°07מ‬C. At ‫°02מ‬C brains were sectioned (20 µm) in a cryostat with alternating sections mounted and exposed for 5 d with standards ( 14 C methylmethacrylate standard 10 ‫ן‬ 0.02 mCi; American Radiolabeled Chemicals, Inc.) (DiRocco and Hall 1981;Coopersmith and Leon 1986;Nudo and Masterton 1986;Sullivan and Wilson 1995). Analysis was done by an experimenter blind to the experiment conditions using the NIH computer-based digital image system that allows pseudocolor imaging and quantitative optical densitometry of autoradiographs.…”
