Metabolic sensitivity of pancreatic tumour cell apoptosis to glycogen phosphorylase inhibitor treatment

Abstract: Inhibitors of glycogen breakdown regulate glucose homeostasis by limiting glucose production in diabetes. Here we demonstrate that restrained glycogen breakdown also inhibits cancer cell proliferation and induces apoptosis through limiting glucose oxidation, as well as nucleic acid and de novo fatty acid synthesis. Increasing doses (50–100 μ) of the glycogen phosphorylase inhibitor CP-320626 inhibited [1,2-13C2]glucose stable isotope substrate re-distribution among glycolysis, pentose and de novo fatty acid sy… Show more

“…4, spermine-exposed VSMC treated with 200 g/ml F1 retained the glycolytic and transamination function and the incorporation of 13 C in alanine commensurate with control VSMC. The addition of NAC to spermine did not restore incorporation of 13 C from glucose to alanine (Fig. 4).…”

“…]glucose (Cambridge Isotope, Andover, MA) at the concentration of 1 g/l for 24 h. At the end of the incubations, cells were separated by centrifugation (1,500 rpm for 5 min), and incubation medium and cell pellets were obtained for tracer-based metabolic studies in conjunction with gas chromatography/mass spectrometry (GC/MS) analysis (3,13).…”

“…Because glucose passes through membrane by facilitated diffusion, with the concentration of glucose increasing obviously, the intracellular glucose may still increase. The increase of intracellular glucose may protect cell [12][13][14] . In contrast, the decrease of intracellular glucose sensitized tumor cells to apoptosis [15,16] .…”
