Polyphenols are a class of plant secondary metabolites that are recently drawing a special interest because of their broad spectrum of pharmacological effects. As they are characterized by an enormous structural variability, the identification of these molecules in food samples is a difficult task, and sometimes having only a limited number of commercially available reference materials is not of great help. One-dimensional liquid chromatography is the most widely applied analytical approach for their analysis. In particular, the hyphenation of liquid chromatography to mass spectrometry has come to play an influential role by allowing relatively fast tentative identification and accurate quantification of polyphenolic compounds at trace levels in vegetable media. However, when dealing with very complex real-world food samples, a single separation system often does not provide sufficient resolving power for attaining rewarding results. Comprehensive two-dimensional liquid chromatography is a technique of great analytical impact, since it offers much higher peak capacities than separations in a single dimension. In the present review, we describe applications in the field of comprehensive two-dimensional liquid chromatography for polyphenol analysis in real-world food samples. Comprehensive two-dimensional liquid chromatography applications to nonfood matrices fall outside the scope of the current report and will not be discussed.

Food chemistry is a branch of food science involved in the evaluation of the composition along with chemical, physical, and functional properties of foods and food products during their different processing stages and storage periods. Chromatographic methods are the most employed analytical techniques for their analysis. However, when a single chromatographic method is not sufficient for providing satisfactory results, more advanced and performant strategies need to be considered. In this regard, comprehensive two‐dimensional chromatography, involving two different separation modes, turned out to be a powerful analytical strategy for the analysis of very complex food samples. This article aims to highlight how the hyphenation of two single separation systems could be a valuable tool for handling such complex samples. The first part deals with general considerations about the technique including the design of modulation interfaces, whereas in the second part selected applications to food analytes of interest, analyzed by both comprehensive two‐dimensional liquid and gas chromatography, are reported and discussed.

Flavonoids are one of the largest groups of plant secondary metabolites. They comprise several thousand compounds that share a phenylchromane skeleton and can be classified into different classes, namely flavones, flavonols, flavanones, flavanols, anthocyanins, dihydroflavonols, isoflavones, and chalcones. Flavonoids occur in their natural sources as aglycones or glycosylated forms and as monomers or constituting polymerized structures and can be found both as free and matrix‐bound compounds. This structural diversity affects their physicochemical behavior, and different flavonoid classes and compounds may have different requisites for their extraction and analysis, so that there is not a unique analytical strategy that applies in all situations. In this article, the main methodological approaches to the analysis of flavonoids in plant materials are revised. Particular attention is paid to more recent extraction techniques and high‐performance liquid chromatography (LC)‐based methodologies coupled to different detection systems, and especially liquid chromatography coupled to mass spectrometry (LC‐MS) that currently dominates the field of flavonoid analysis.
