Metachronal activity of cultured mucociliary epithelium under normal and stimulated conditions

Abstract: In the present work we measured in real time the metachronism and degree of correlation between beating cilia from cultured mucociliary epithelium. The method is based on simultaneous measurement of ciliary beat frequency, phase shifts, and correlation factors in two directions: parallel and perpendicular to the effective stroke direction (ESD). From the phase shifts the lengths of wave components, and consequently the metachronal wavelength and direction, were evaluated. On active ciliary areas of cultured fr… Show more

“…At the moment this is a speculation consistent with a number of experimental observations (9,11,15,21). The characteristic delay time we have found should not be generally present in stochastic synchronizing systems where the coupling does not induce the same correlation properties in the noise (31).…”

“…cilia are stable, so that robust mechanisms to induce them must exist. Cilia play vital roles on the surface of the respiratory tract in mammals, maintaining an upward flow of mucus, away from the lungs (9). They also determine the asymmetry of various organisms during development (10).…”

“…In the simulations, it is found to be roughly parallel to the powerstroke direction, see Fig. 3, in agreement with observations for frog esophagus (35). The main correlation direction depends very weakly on cilia spacing; some small systematic deviation can be seen in Fig.…”

“…We find that the wave length is about two cilia lengths, λ ' 2L c , depending only weakly on both d c and Θ. This agrees well with experiments on frog esophagus (35). It is important to realize that the propagation direction of the MCW does not have to be the same as the power-stroke direction.…”

“…The whole field of 640 by 480 pixels was divided into equal sized blocks (spots) of 2 by 2 pixels. Gray-level values of all pixels in each spot were summed sequentially from the first frame to the last.Movements of cilia are coordinated in time and space by phase differences, and this is called metachronism or a metachronal wave [10]. The Pearson correlation factor between series from two different spots determined the degree of synchronization between ciliary beats.…”
