Metal Ion Trafficking in Earthworms

Abstract: Exposure to cadmium poses a considerable risk to human health and environmental safety. Earthworms reside in the most contaminated sites on earth, displaying a phenomenal tolerance to toxic heavy metals. They exhibit a distinct metabolic pathway that allows the bio-accumulation of cadmium to yield body burdens in excess of 1/1000th of total dry body weight, a most impressive figure by any standard. However, the precise molecular mechanism underlying this phenomenon remains to be unraveled. This study meets thi… Show more

“…There are 33 cysteine residues present and no disulfide bonds. In addition to the sequences from bbbhMT and aaahMT, the expression systems include the amino acid residues of the stabilizing S-peptide tag (MKETAAAKFERQHMDSPDLGTLVPRGS) on the N-terminus of each protein [40,41]. Recombinant bbbhMT-1a and aaahMT-1a were expressed and purified as previously reported [20,21].…”

“…The rate constants for the As 3+ -metalation are reported for both the bbb and aaa human MT. At room temperature (298 K) and pH 3.5, the sequential individual rate constants, k n (n = 1-9) for the As 3+ -metalation of bbbhMT starting at k 1bbb are 40,36,37,26,27,17,12,6, and 1 M À1 s À1 ; while at room temperature (298 K) and pH 3.5, the sequential individual rate constants, k n (n = 1-10) for the As 3+ -metalation of aaahMT starting at k 1aaa are 52, 45,46,42,38,36,29,25,14, and 6 M À1 s À1 . The trend in the rate constant values reported for these two triple-domain MT proteins supports our previous proposal that the rate constant values are proportionally related to the total number of equivalent binding sites.…”

“…Unlike E. fetida , two MT isoforms, displaying an identity of 74.7% and a similarity of 91.1%, have been isolated in L. rubellus . All cysteine residues are conserved between the two isoforms, but interestingly there is a distinctive two amino acid deletion/insertion [31]. Whereas Oligochaeta are used to monitor terrestrial heavy metal pollution, Polychaeta could be useful bioindicators for assessing coastal marine pollution, but few studies have investigated MTs in these invertebrates [32].…”

“…Three Cd-binding MT isoforms have been found in the earthworm, Lumbricus rubellus (Stürzenbaum et al, 1998, of which only the wmt2 isoform is Cd-inducible (Stürzenbaum et al, 2001); wmt3 is enriched in embryonic tissue . The genomic organisation of the isoforms is complicated.…”
