Metalloproteinase/Presenilin1 processing of ephrinB regulates EphB-induced Src phosphorylation and signaling

Abstract: Bidirectional signaling triggered by interacting ephrinB receptors (EphB) and ephrinB ligands is crucial for development and function of the vascular and nervous systems. A signaling cascade triggered by this interaction involves activation of Src kinase and phosphorylation of ephrinB. The mechanism, however, by which EphB activates Src in the ephrinB-expressing cells is unknown. Here we show that EphB stimulates a metalloproteinase cleavage of ephrinB2, producing a carboxy-terminal fragment that is further pr… Show more

“…[3][4][5][6][7] In general, the EPH kinases interact with their EFN ligands on neighboring cells, because EPHs and EFNs are all cell surface molecules. 2 These molecules could be cleaved from the cell surface by enzymes such as ADAM10, 8,9 an unspecified matrix metalloproteinase, 10 or γ-secretase; 11 therefore, it is possible that the shed soluble fragments of EPH and EFN might be able to influence cells and tissues at a distance by blocking the interaction of EPHs and EFNs there.…”

“…As well as triggering RTK activity in the receptor-bearing cells, a signal is triggered in the ligand-expressing cell (Georgakopoulos et al 2006). This is important for the role of ephrinB2 in cardiac valve maturation and axon path finding (Cowan et al 2004).…”

“…Several mechanisms are known to regulate the tyrosine phosphorylation of ephrinBs. First, EphB receptor stimulates a metalloproteinase cleavage of ephrinB ligands, producing a C-terminal fragment that is further processed by presenilin 1/γ-secretase [28,80]. The final peptide product binds Src and inhibits its association with a negative Src regulator termed cterminal Src kinase (Csk), allowing further activation of Src [28].…”
