Metallothioneins and zinc in cancer diagnosis and therapy

Abstract: Metallothioneins (MTs) are involved in protection against oxidative stress (OS) and toxic metals and they participate in zinc metabolism and its homeostasis. Disturbing of zinc homeostasis can lead to formation of reactive oxygen species, which can result in OS causing alterations in immunity, aging, and civilization diseases, but also in cancer development. It is not surprising that altered zinc metabolism and expression of MTs are of great interest in the case of studying of oncogenesis and cancer prognosis.… Show more

“…4, all four measured indicators amounted to the highest value in fibropapillomas. The role of MTs and zinc in cancer development is closely linked and the structure and function of MTs depend strongly on the redox state of zinc and its interactions with proteins (Krizkova et al 2012). A similar increase in the concentration of MTs has previously been documented in skin melanoma cells both in vivo and in vitro (Krizkova et al 2008).…”

“…At the protein level, MT has been examined as a marker of cancer [29] and also of metals' and toxic compounds' intoxication [30,31]. For these purposes, numerous analytical and biochemical methods are used [32,33], whereas the electrochemical ones seem to be the most sensitive [33][34][35].…”

“…MTs are widely distributed in invertebrates and vertebrates, plants, prokaryotes and even in fungi kingdoms [4][5][6][7]. Despite the diversity of the amino acid sequences, the MTs are generally analogous in structure with respect to a high content of cysteine residues (up to 30%), and lack of aromatic amino acids [4].…”

“…Despite the diversity of the amino acid sequences, the MTs are generally analogous in structure with respect to a high content of cysteine residues (up to 30%), and lack of aromatic amino acids [4]. MTs are involved in the metabolism of heavy metal ions, including their metal detoxification, homeostasis [8,9], storage of zinc [7], radical scavenging [2,10] and stress response [11]. MTs are overexpressed in several tumors and this overexpression is accompanied by an increased proliferation and protection against apoptosis.…”
