Metformin Is a Substrate and Inhibitor of the Human Thiamine Transporter, THTR-2 (SLC19A3)

Abstract: The biguanide metformin is widely used as first-line therapy for the treatment of type 2 diabetes. Predominately a cation at physiological pH’s, metformin is transported by membrane transporters, which play major roles in its absorption and disposition. Recently, our laboratory demonstrated that organic cation transporter 1, OCT1, the major hepatic uptake transporter for metformin, was also the primary hepatic uptake transporter for thiamine, vitamin B1. In this study, we tested the reverse, i.e., that metform… Show more

“…Growth differentiation factor 15 ( GDF15 ) was the top upregulated AMPK-independent gene (Fig 2B) and its overexpression is known to result in improved insulin sensitivity [50]. The transporter solute carrier family 19, member 3 ( SLC19A3 ) was also an upregulated AMPK-dependent gene (S1 Table), and is known to play a role in the intestinal absorption and tissue distribution of metformin [51]. These findings further support that metformin induces protein coding genes which play a role in cancer and T2D.…”

“…The transcript levels of SLC2A2 were determined using TaqMan probe (ID Hs01096908_m1). The relative expression level of SLC2A2 transcript was calculated by the comparative method (ΔΔCt) normalized to the housekeeping gene GAPDH, as described previously52.…”

“…This led to the discovery that the antibiotic trimethoprim also potently inhibits thiamine uptake [272]. Metformin is also both a substrate and an inhibitor of one of the transporters [275].…”
