Vaporizing drugs in e-cigarettes is becoming a common method of administration for synthetic cathinones and classical stimulants. Heating during vaporization can expose the user to a cocktail of parent compound and thermolytic degradants, which could lead to different toxicological and pharmacological effects compared to ingesting the parent compound alone via injection or nasal inhalation. This study examined the in vivo toxicological and pharmacological effects of vaporized and injected methamphetamine (METH) and α-pyrrolidinopentiophenone (α-PVP). Male and female ICR mice were administered METH or α-PVP through vapor or i.p. injection. Dose-effect curves were determined for locomotor activity and a functional observational battery (FOB). METH and α-PVP vapor were also evaluated for place preference in male mice. Vapor exposure and injection led to more similarities than differences in toxicological and pharmacological effects. In the FOB, both routes of administration produced typical stimulant effects, and injection also increased some bizarre behaviors (e.g. licking, teeth chattering, darting). Both METH and α-PVP vapor exposure produced conditioned place preference. The two routes of administration had comparable efficacy in locomotor activation, with vapor producing longer lasting effects than injection. Females showed greater METH-induced locomotor activity, and greater incidence of a few somatic signs in the FOB than males. These results explore the toxicology of stimulant vapor inhalation in mice using an e-cigarette device. Despite the current technological and methodological difficulties, studying drug vapor promises to allow determination of toxicological effects of thermolytic products and flavor additives.

Accumbal 14-3-3ζ Protein Downregulation Is Associated with Cocaine-Conditioned Memory

Cocaine-conditioned memory has been known to cause cocaine craving and relapse, while its underlying mechanisms remain unclear. We explored accumbal protein candidates responsible for a cocaine-conditioned memory, cocaine-induced conditioned place preference (CPP). Two-dimensional gel electrophoresis in conjunction with liquid chromatography mass spectrometry analysis was utilized to identify accumbal protein candidates involved in the retrieval of cocaine-induced CPP. Among the identified candidate proteins, a downregulated 14-3-3ζ protein was chosen and confirmed by Western immunoblotting. A polymer-mediated plasmid DNA delivery system was then used to overexpress 14-3-3 protein in mouse nucleus accumbens before the CPP retrieval tests. Overexpression of accumbal 14-3-3ζ protein was found to diminish conditioned cue/context-mediated cocaine-induced CPP. In contrast, another isoform of 14-3-3 protein, 14-3-3ε protein, did not affect conditioned cue/context-mediated cocaine-induced CPP. Overexpression of accumbal 14-3-3ζ protein did not produce motor activity-impairing effect or alter local dopamine metabolism. Moreover, overexpression of accumbal 14-3-3ζ protein did not affect food-induced CPP. These results, taken together, indicated that overexpressed accumbal 14-3-3ζ protein specifically decreased conditioned cue/context-mediated cocaine memory. Further understanding of the function of accumbal 14-3-3ζ protein may shed light on the treatment of cocaine craving and relapse.

bstractObjectives: A robust stressor regimen may cause a rapid decrease in BDNF level in the dentate gyrus (DG) while social buffering appears to prevent such decrease. Local BDNF levels and downstream phosphorylation of ERK and CREB play a critical role in mediating morphological and synaptic plasticity in DG. Thus, this study was undertaken to assess whether an acute stressor regimen may render morphological changes and whether the social buffering may prevent such changes in the existing DG granule cells. Moreover, we attempted to assess whether the stressor regimen and social buffering may also affect local ERK and CREB phosphorylation and the hippocampus-related memory.Methods: Six hours after the conclusion of the stressor regimen, morphological indices of the granule cell in DG were obtained in Balb/C mice experiencing no stressor, the stressor regimen alone or in a group. Results:The stressor caused significant decreases in the total length of dendrites, number of dendritic branches, and the size of the dendritic field in granule cells, while the social buffering prevented all these changes. Likewise, the stressor caused decreases in local ERK phosphorylation and object location performance, while the social buffering prevented such decreases of ERK phosphorylation and deteriorated memory performance. Conclusion:These results, taken together, suggest that stress and social buffering may rapidly affect the existing DG granule cell morphology and hippocampus-related memory performance by modulating local ERK phosphorylation.
