Methylome Analysis in Chickens Immunized with Infectious Laryngotracheitis Vaccine

Abstract: In this study we investigated the methylome of chickens immunized with Infectious laryngotracheitis (ILT) vaccine derived from chicken embryos. Methyl-CpG binding domain protein-enriched genome sequencing (MBD-Seq) method was employed in the detection of the 1,155 differentially methylated regions (DMRs) across the entire genome. After validation, we ascertained the genomic DMRs distribution and annotated them regarding genes, transcription start sites (TSS) and CpG islands. We found that global DNA methylatio… Show more

“…This situation also resembles that in chickens insofar as the DNA methylome of resistant chickens responds to MDV infections with more downthan up-methylations in comparison to susceptible chicken (Tian et al 2013). Furthermore, chickens, when immunized with an infectious laryngotracheitis vaccine, have been recently shown to exhibit massive changes in global DNA methylation with more down-methylations than non-vaccinated control chicken (Carillo et al 2015). Though the previous studies with chickens are not completely comparable with our studies in mice, both studies have at least in common that survival, and even the potency for survival, of two different infectious diseases in two different hosts is associated with more downthan up-methylations of host DNA.…”

“…“EIF2 signaling” is also predicted to have higher activity in trachea of Fayoumis compared to Leghorns at 2 dpi 13 , and it has been known to inhibit viral replication and regulate proinflammatory cytokine expression 36 . Interestingly, all three pathways were reported to show decreased methylation in chickens immunized with infectious laryngotracheitis vaccine compared to the unimmunized ones 37 . Therefore, upregulation of these pathways may be vitally important for enhanced disease resistance in Fayoumi chickens.…”

“…Genome-wide DNA methylation of lowland chickens has been investigated using MeDIP-seq [ 19 , 20 ], MBD-Seq [ 21 ], and Methyl-MAPS [ 22 ], which measure methylation based on immunoprecipitation and restriction enzyme digestion. Compared to WGBS, these technologies generate lower resolution and coverage and fail to obtain methylation level for CHG and CHH.…”
