Micafungin-Induced Suicidal Erythrocyte Death

Abstract: Background/Aims: The antifungal drug Micafungin is used for the treatment of diverse fungal infections including candidiasis and aspergillosis. Side effects of Micafungin treatment include microangiopathic hemolytic anemia and thrombocytopenia with microvascular thrombosis. The development of thrombosis may be fostered by stimulation of eryptosis, the suicidal death of erythrocytes characterized by cell shrinkage and cell membrane scrambling with phosphatidylserine translocation to the erythrocyte surface. Tri… Show more

“…7,9) Nonetheless, our data show that PSN-induced eryptosis is not mediated through oxidative stress (Fig. 6), unlike emodin, 21) but similar to micafungin, 15) triclosan, 25) and N,N-diethyl-3-methylbenzamide. 26) Therefore, counteracting eryptosis by antioxidants is unlikely to attenuate PSN-induced cell death.…”

“…13) Circulating hemoglobin causes oxidative damage to vital organs, and aggravates renal disease. 15) Moreover, hemoglobin degradation products interfere with immune function and exacerbate inflammatory conditions. 16,17) PSN also induced significant loss of membrane asymmetry (Fig.…”

“…50 Another recent report claimed that MCFG induces hemolysis by triggering cell shrinkage and cell membrane scrambling in human erythrocytes. 51 Although further investigations on the effects of MCFG to mouse blood are warranted, we suspect that this effect may be related to a similar mechanism as proposed in humans. We also observed spleen enlargement in the MCFG-treated mice; although the spleen expansion was only reduced by 0.5-fold compared with that of the DENV-infected group, it was significantly (p < 0.05) enlarged relative to that of untreated healthy mice.…”

“…In humans, the drug label indicates that high dosing could induce hemolysis, which is supported by a clinical report . “…Erythrocytes from newborns are highly sensitive to oxidative stress leading to rapid eryptosis and subsequent clearance of those erythrocytes following birth . Eryptosis is triggered by complement, hyperosmotic shock, energy depletion, oxidative stress, cellular K + loss, hyperthermia and a wide variety of xenobiotics including diverse cytostatic drugs (see below).…”
