Mice Lacking beta2-Integrin Function Remain Glucose Tolerant in Spite of Insulin Resistance, Neutrophil Infiltration and Inflammation

Abstract: Beta2-integrins are important in leukocyte trafficking and function, and are regulated through the binding of cytoplasmic proteins, such as kindlin-3, to their intracellular domain. Here, we investigate the involvement of beta2-integrins in the regulation of metabolic disease using mice where the kindlin-3 binding site in the beta2-integrin cytoplasmic tail has been mutated (TTT/AAA-beta2-integrin knock-in (KI) mice), leading to expressed but dysfunctional beta2-integrins and significant neutrophilia in vivo. … Show more

“…They had about 10-fold increased serum IgG levels, and elevated IL-3 and IL-6 serum levels as compared with WT mice. The CD18 −/− mouse model resembles therefore a severe form of the LAD-I syndrome in humans and has been explored in the context of various pathologies like psoriasis [180], wound healing [181], diabetes [182], carditis [183,184], osteoporosis [185], and infections [186].…”

“…These cells transiently infiltrate murine adipose tissue with the initiation of high-fat-diet feeding and mediate insulin resistance in mice. 24 , 25 Natural killer (NK) cells, best known for their role in the early host response to viral infections and in killing tumor cells, have recently been shown to promote macrophage proliferation and polarization in adipose tissue and insulin resistance in mice fed a high-fat diet. 26 …”

“…On the other hand, leucocyte-derived β2 integrin has been associated with HFD-induced obesity and insulin resistance in the adipose tissue. Under a HFD condition, mutated β2-integrin knockin mice display increased neutrophil numbers in white adipose tissues and show significantly increased peripheral insulin resistance [72]. The β2 integrin subfamily is comprised of 4 members, αLβ2 (CD11a/CD18), αMβ2 (CD11b/CD18), αXβ2 (CD11c/CD18), and αDβ2 (CD11d/CD18).…”
