Microbial Bioprocess for Extracellular Squalene Production and Formulation of Nanoemulsions

Abstract: Sustainable production of natural resources requires a clean manufacturing process with clean raw materials due to the risk of environmental contaminations. Microbial cell factories have enabled the production of natural products including hydrophobic chemicals. However, the microbial cells limit the production titer because of the limited intracellular storage capacity and no transport across the cell membrane. Herein, we engineered Corynebacterium glutamicum to produce hydrophobic squalene as a model product… Show more

“…glutamicum strains using pHsSQS-DIIDF. To improve squalene production, the native decaprenoxanthin biosynthesis has been blocked by mutating idsA ::E16Ter and crtE ::D21Ter to redirect carbon flux toward squalene biosynthesis. , Thus, DM1919 was engineered by the same strategy, and a strain DM1919-Δcrt was constructed. The squalene production of DM1919-Δcrt pHsSQS-DIIDF (5.37 ± 0.51 g/L l -lysine and 118.15 ± 3.30 mg/L) resulted in 1.7-fold production of DM1919 pHsSQS-DIIDF (4.90 ± 0.13 g/L l -lysine and 69.49 ± 4.02 mg/L) (Figure C).…”
