Microbial Warfare on Three Fronts: Mixed Biofilm of Aspergillus fumigatus and Staphylococcus aureus on Primary Cultures of Human Limbo-Corneal Fibroblasts

Abstract: BackgroundCoinfections with fungi and bacteria in ocular pathologies are increasing at an alarming rate. Two of the main etiologic agents of infections on the corneal surface, such as Aspergillus fumigatus and Staphylococcus aureus, can form a biofilm. However, mixed fungal–bacterial biofilms are rarely reported in ocular infections. The implementation of cell cultures as a study model related to biofilm microbial keratitis will allow understanding the pathogenesis in the cornea. The cornea maintains a pathoge… Show more

“…This data was evaluated by the respective statistic methods described in the following paragraphs. To quantify the bio lm developed by determining its metabolic activity, the technique described by Walencka et al [16], modi ed by Ramírez-Granillo et al, was used [15,57]. The bio lm was developed in 96-well polystyrene plates with a at bottom, as described previously.…”

“…Bacterial contamination of the prosthetic components often occurs during their implantation. Despite the wide acceptance of the seminal concept of 'race for the surface' between the bacteria and host, recent findings indicate that the severity of the resulting infection largely depends on the bacterial strain, bacterial load, and the environment [42][43][44][45][46][47]. In our study, the ATCC 12600 strain was selected to simulate an infection that is susceptible to commonly used antibiotics (low risk) in the clinic.…”

“…Filamentous fungi harbor diverse adaptive mechanisms encoded in their genome, facilitating their pathogenic behavior in the human host [13,14]. The formation of biofilms represents a virulence factor that supports the creation and persistence of infections [15]. Concern about the biofilms formed by filamentous fungi extends beyond the healthcare system to the food industry.…”
