Microbiological assay for vitamin B12 performed in 96-well microtitre plates.

Abstract: A simplified microbiological assay for vitamin B12 estimation, completed on microtitre plates using a colistin sulphate resistant strain of Lactobacillus leichmannii (NCIB 12519), and cryopreserved cultures is described. The new assay correlated well with a more conventional "tube" assay and was not influenced by the presence of antibiotics in serum. Evaluation of assay performance showed excellent interassay and intra-assay precision with quantitative recovery of added cyanocobalamin over a wide range of addi… Show more

“…All assays were done by laboratory personnel who were blinded as to the case-control status of the blood samples. Folate concentrations were determined by a Lactobacillus casei microbiological assay (13) and vitamin B 12 concentrations were determined by a Lactobacillus leichmannii microbiological assay (14,15). Both folate and vitamin B 12 assays were adapted to a microtiter plate format and carried out by a robotic workstation (Microlab AT plus 2; Hamilton Bonaduz).…”

“…23 Folate and vitamin B12 concentrations were determined with a microbiological method using Lactobacillus casei and Lactobacillus leichmannii, respectively, and adapted to a microtiter plate format and carried out by a robotic workstation. 24,25 in addition, three single nucleotide polymorphisms (BHMT 742g>a, MTR 2756a>g, and MTHFR 677c>t) were determined using MalDi-tOF mass spectrometry. 22 to prevent batch effects, samples were analyzed in matched case-control sets, with the position of the case randomized within each set.…”

“…Serum concentrations of folate and vitamin B12 (cobalamin) were measured by microbiological assays (30,31), whereas methionine, choline, betaine, vitamin B2 species [riboflavin and flavin-mononucleotide (FMN)], and vitamin B6 species [pyridoxal 5 0 -phosphate (PLP), pyridoxal (PL), and 4-pyridoxic acid (PA)] were measured in plasma using liquid chromatography-tandem mass spectrometry (32,33). Samples were analyzed in batches of 86 and quality control included six calibration samples, two Figure 1.…”
