Lactic acid bacteria can convert selenium (Se) from inorganic to organic and elemental forms, but the distribution and existence form of organic Se in the bacteria are not clear after Se enrichment, and the effects of selenization on the growth and nutritional value of strains also need to be studied. In this study, Pediococcus acidilactici MRS-7 could absorb up to 67% of inorganic Se and convert most of it into organic Se; about 75% of organic Se was selenoprotein, 2.7% was Se-polysaccharide, and 4.6% was Se-nucleic acid. Additionally, Se-enriched treatment increased the levels of amino acids and essential elements in P. acidilactici MRS-7. Finally, after Se enrichment, Se nanoparticles (SeNPs) were found on the surface of P. acidilactici MRS-7, but they had no harmful effect on its morphology, and its survival during gastrointestinal digestion was not affected, indicating that SeP has potential probiotic value in the food industry.

Probiotics, microorganisms that can benefit animal or human hosts, face application challenges due to environmental factors that reduce their stability. However, techniques like microencapsulation improve survival rates. Additionally, probiotics possess antibacterial properties that can inhibit the growth of pathogenic bacteria accountable for foodborne illnesses. The aim of this study was to assess the viability of microencapsulated Lactobacillus plantarum under simulated gastrointestinal conditions and its potential probiotic impact on Campylobacter jejuni. Fermentation kinetics was evaluated in an MRS culture medium over 24 hours. The growth of L. plantarum at 37°C and 45°C was examined, as well as microencapsulation through spray drying. Additionally, exposure to simulated gastrointestinal conditions was analyzed, while inhibition tests of L. plantarum on C. jejuni were performed. Finally, exopolysaccharide production from L. plantarum was assessed. The study findings demonstrated the termination of the exponential growth phase after 15 hours, improved development of lactic bacteria at 37°C, microencapsulation parameters within acceptable limits, survival of the microencapsulated strain in in vitro gastrointestinal conditions exceeding 7x108 UFC, and significant inhibitory effects of L. plantarum on pathogenic bacteria. The viability of microencapsulated Lactobacillus plantarum, subjected to simulated gastrointestinal conditions, exceeded 7x108 CFU/mL and demonstrated a probiotic effect on Campylobacter jejuni.

To improve the survivability of probiotics, Lactobacillus plantarum was microencapsulated using pufferfish skin gelatin (PSG)-based wall materials by spray-drying. This work investigated the protective effect of three different pH-dependent proteins (sodium caseinate (SC), soy protein isolate (SPI), and whey protein isolate (WPI)) combined with PSG on L. plantarum. The experimental results of spray-drying with an inlet temperature of 120 °C and an outlet temperature of 80 °C, storage at 4 °C for 6 months, simulated digestion, and turbidity indicated that PSG/SC had better stability and encapsulation effects and was more suitable to encapsulate L. plantarum than PSG/SPI and PSG/WPI. The optimum preparation conditions for L. plantarum microcapsules were a PSG/SC mass ratio of 2:1, an SC concentration of 20 g/L, and a cell concentration of 10 g/L. The encapsulation efficiency of the obtained microcapsules was 95.0%, and the survival rate was 94.2% in simulated gastric fluid for 2 h and 98.0% in simulated intestinal fluid for 2 h. Amino acid composition analysis exhibited that the imino acid and aspartic acid contents of PSG were 27.98 and 26.16 g/100 g protein, respectively, which was much higher than commercial bovine gelatin. This characteristic was favorable to the high encapsulation efficiency and stability of microcapsules. In vitro release experiments showed that the PSG/SC microcapsules did not disintegrate in simulated gastric fluid for 2 h but could completely release in simulated intestinal fluid for 2 h, which can maintain the high survivability of L. plantarum in simulated digestion. In general, this study demonstrated that microcapsules using PSG/SC as wall materials can effectively improve the survivability of probiotics and have great potential for application in probiotic products.
