Microfluidics-based point-of-care test for serodiagnosis of Lyme Disease

Abstract: Currently, diagnostic testing for Lyme disease is done by determination of the serologic responses to Borrelia burgdorferi antigens, with the exception of the early localized phase of disease where diagnosis must be done clinically. Here, we describe the use of microfluidics technology to develop a multiplexed rapid lab-on-a-chip point of care (POC) assay for the serologic diagnosis of human Lyme disease. Following ELISA screening of 12 candidate antigens, we tested 8 on a microfluidic diagnostic system, calle… Show more

“…Point‐of‐care (PoC) diagnostics have recently emerged as an analytical alternative for remote settings. Microfluidic devices are attractive candidates for use with PoC applications for diagnostics . Alternatively, a PoC device could detect ThDP (or chemical analogs) based on electrochemical properties.…”

“…A recent form factor was a smartphone accessory 216 that operated on disposable low-cost microfluidic cassettes (about $1.50 in material cost per chip) which consisted only of plastic and chemical reagents. Recently, we have also demonstrated integration of two different classes of diagnostic tests into this single platform, showing simultaneous measurement of hemoglobin concentration and HIV antibody detection 217 , and detection of Lyme disease 279 .…”

“…In developing countries where illiteracy is more common, there is a dire need for fully automated onsite disease‐diagnostic devices that can perform complex laboratory assays in a simplified way at low costs . One such microfluidic POC test device with integrated CMOS‐based photosensors, demonstrated by Nayak et al, was capable of detecting Lyme disease . Lyme antigens (toxins that induce an immune response in the body) from a patient's blood sample were immobilized on a microfluidic cassette, then exposed to antibodies and silver amplification reagents, which initiated a sequential binding.…”

“…This binding caused a decrease in the light from an LED that reached a photodiode at the top of the device, thus indicating the presence of Lyme antigens. An illustration of the detection system can be seen in Figure a …”
