Microneutralization test for respiratory syncytial virus based on an enzyme immunoassay

Abstract: Virus infectivity and antibody neutralization titers for respiratory syncytial virus were determined in cell cultures in microtiter plates. After an appropriate incubation period, the cells were fixed, and an enzymelinked immunosorbent assay was performed directly in the microtiter plates for detection of virus. Results could be read and recorded automatically, which is especially helpful when running large numbers of tests.

“…Infected cells were scraped into the culture medium when viral cytopathic effect involved the whole monolayer and cell suspensions were stored at À708C in 0.5 ml aliquots. The thawed suspensions were titrated by a fluorescent focus assay as described by Anderson et al [1985].…”

“…Microneutralization assays were performed using a modification of the method previously reported by Anderson et al [1985]. Serial log, dilutions of each serum sample (1:50 to 1:256,000) in MEM were incubated with approximately 100 plaque forming units of RSV (Long strain and 18537).…”

“…The anti-CMV reactivity of the IgG fractions was independent of pH treatment. In an RSV-micro neutralization assay [29], IgG and its fractions decreased the degree of viral infection of the susceptible HEp-2 cells in a dose-dependent manner and with comparable efficacy (Fig. 1B).…”
