The procedure of micropropagation for efficient and stable conservation of Monarda fistulosa L., a medicinally important plant belonging to the Lamiaceae family, was developed. Our research consisted of the introduction in vitro culture of the precious genotype from a biochemical point of view, identified as a result of multiple analyzes carried out by the researchers of the Scientific Medicine Research Center of the ʼNicolae Testemitanuʼ State University of Medicine and Pharmacy. This genotype is characterized by an increased content of secondary metabolites (thymol and carvacrol). Axillary bud proliferation was initiated from nodal explants grown on ½ Murashige-Skoog (MS) nutrient medium supplemented with various cytokinins, such as 6-benzylaminopurine (BA), 6-(α, α-dimethylallylamino)-purine (2iP) and cytokinin-like growth regulators -thidiazuron (TDZ) with a concentration of 0.5 mg.L -1 . After 6 weeks of culture on a nutrient medium, containing 0.5 mg.L -1 BA was determined the maximum number of axillary shoots per explant (2.75 ±0.86) and the highest number of internodes per shoot (11.66 ±1.30). At the same time, the rhizogenesis was induced on that medium that allows in vitro micropropagation by a single stage. The advantages of the elaborated procedure are reducing reagents and energy expenses for in vitro cultivation by two times and increasing the coefficient of multiplication from 13 to 32.

From an ornamental viewpoint, tulips are famous clonally propagated crops. This research focuses on 15 wild-growing Greek tulip species including 11 range-restricted species, i.e., six Greek endemics and five Balkan or Aegean endemics and subendemics, among which seven are currently threatened with extinction (two Critically Endangered, three Endangered and two Vulnerable). An overview of the global electronic trade over the internet is presented herein for these valuable phytogenetic resources in an attempt to define the extent of their commercialization (25 nurseries in three countries, mainly bulb trade at various prices) with concomitant conservation implications. In the frame of the repatriation initiatives launched, their global ex situ conservation is overviewed according to the PlantSearch facility of the Botanic Gardens Conservation International (materials from 15 species stored in 41 botanic gardens of 14 countries). The results of this study on the Greek tulips showed that there are both well-established value chains and gaps in the market regarding the “botanical tulips”; revealed the compromised effectiveness of ex situ conservation for the majority of them; raised conservation concerns related to authorized access to these wild phytogenetic resources; and indicated that their future utilization should comply with the provision of national and international legislation. All these are envisaged and discussed within the framework of the newly launched research project TULIPS.GR which aims to be the pilot establishment of a national collection regarding all Greek tulips (currently holding 38 accessions of 13 species, including almost all of the threatened ones). The project’s scope is to enable the creation of a sustainable value chain for the Greek tulips with authorized collections, sustainable conservation schemes, production of DNA barcoded propagation material, species-specific propagation and cultivation protocols, mycorrhizal investigations, field studies, applying innovative precise soil/foliar fertigation, and investigation of the postharvest treatment of fresh cut flowers, promoting networking and synergies with producers and associations in Greece and abroad.

The aim of the work was to study the influence of cultivation factors on the explants development to optimize the protocol of in vitro clonal micropropagation of four species of the genus Thymus L., valuable essential oil and medicinal plants. Conditions that ensure the production of up to 94.2 % of sterile explants were selected. It has been established that, when introduced into in vitro culture, it is advisable to use shoot tips and stem segments with a node (isolated from donor plants) and cultivate them in foil-covered test tubes. The optimal culture media for the first stage of micropropagation were determined: Murashige and Skoog (MS) medium with 1.0 mg/l benzylaminopurine (BAP) for Thymus serpyllum L. or MS with 1.0 mg/l kinetin and 1.0 mg/l gibberellic acids (GA3) for T. vulgaris L. ‘Gornyy Balzam’, T. × citriodorus (Pers.) Schreb. ‘Doone Valley’, T. caucasicus Willd. ex Ronniger. At the second stage of clonal micropropagation, it is advisable to cultivate T. caucasicus and T. serpyllum explants during 40 days, but T. vulgaris and T. × citriodorus – during 60 days. The maximum multiplication index at the recommended cultivation duration was obtained for T. × citriodorus (9.9) on MS medium without hormones, T. caucasicus (16.1) and T. vulgaris (10.0) – on MS medium with 1.0 mg/l kinetin., and T. serpyllum (6.8) on MS medium with 1.0 mg/l BAP. When comparing different culture vessels (tubes, jars and flasks), the effectiveness of jars was revealed. This type of culture vessels made it possible to increase the multiplication index up to 2.1–5.7 times. It was revealed that when microshoots of four thyme species were cultivated on MS medium with 1.0 mg/l IBA, the rooting rate reached 84.6–98.9 %. The frequency of adaptation ex vitro when using a mixture of peat and perlite (1:1) ranged from 62.8 to 89.5 %.
