Micropropagation of Helleborus orientalis Lam. and Aconitum uncinatum Linn. (Ranunulaceae)

Abstract: The objectives of this study were to develop systems for mass proliferation, rooting, and reestablishment of microcuttings of Helleborus orientalis and Aconitum uncinatum. Basal medium for H. orientalis contained 1/2 MS with 0.4 mg thiamineHCl/liter, 0.5 mg pyridoxineHCI/liter, 0.5 mg nicotinic acid/liter, 100 mg i-inositol/liter, 5 mg BA/liter, 2% sucr… Show more

“…For H. niger, a satisfactory survival rate for rooting in vitro (87%) and ex vitro (85%) was obtained after chilling (7°C) microshoots in an IBA and NAA-containing solution for 7 days [18]. A low survival percentage (50-56%) was found for ex vitro rooting of H. orientalis [15]. Dhooghe and van Labeke [17] suggested ex vitro rooting for four Helleborus species which were induced for one week by drenching them in a solution of IBA and NAA at 5°C, but no data on the rooting rate and survival percentage was provided.…”

“…Helleborus species has been investigated in vitro and ex vitro [11,[15][16][17][18]. Studies on in vitro rooting have shown that cultivation of H. niger shoots on IBA and NAA-containing media resulted in a higher proportion of rooted shoots than when cultivation took place on auxin-free medium.…”

“…developed by Lim and Kitto (1995) is suitable for H. niger. The advantage of this system is that the rooting could be carried out ex vitro.…”

“…Springer (1995) reported a successful attempt at in vitro propagation without, however, giving detailed information about either the Helleborus species propagated or the cultivation method used. Lim and Kitto (1995) presented a system used for H. orientalis Lam. in which the propagation of shoots was carried out in vitro and the rooting was done ex vitro.…”
