MicroRNA‑139‑5p elevates skeletal myogenic differentiation of human adult dental pulp stem cells through Wnt/β‑catenin signaling pathway

Abstract: The aim of the present study was to identify a microRNA (miRNA or miR)-based biomarker and therapeutic target for skeletal myogenic differentiation of human adult dental pulp stem cells (ADSCs). miRNA expression was measured using reverse transcription-quantitative polymerase chain reaction; cell viability assay and lactate dehydrogenase (LDH) activity levels were measured using MTT and LDH activity kits, respectively. Apoptosis assay and caspase-3/9 activity levels were measured using flow cytometry and caspa… Show more

“…miR-139-5p was reported to inhibit tumorigenesis and reverses drug resistance in many cancers (Wang et al , 2017 ; Jiang et al , 2018 ; Zhang et al , 2018 ). Targets of miR-139-5p , such as c-Jun (Jiang et al , 2018 ; Su et al , 2018 ), PI3K/Akt (Maoa et al , 2015 ; Catanzaro et al , 2018 ), IGF-1/IGF-1R (Xu et al , 2015 ), VEGF, and Wnt/β-catenin (Long et al , 2017 ; Xie and Shen, 2018 ), are potent stimulators of cell proliferation, some of which are important pathways promoting injury repair (Zhou et al , 2016 ; Guise and Chade, 2018 ; Xie et al , 2018 ). Therefore, we postulate that the downregulation of miR-139-5p may reflect an activation of cell proliferation, promoting the repair and regeneration of injured kidney.…”

“…After being cultured in specific condition for several weeks, DPSCs elongate and display a myoblast-like phenotype. These DPSCs express specific myocytic immunohistochemical markers such as MyoD1, myosin, and MHC [ 79 , 80 ]. Other than the myogenic potential, DPSCs also preserve the capability to differentiate into adipocytes [ 81 , 82 ] and pancreatic cell lineage [ 83 , 84 ].…”

“…The addition of miR-143 or miR-135 inhibitors to culture medium stimulates the myocytic properties of DPSCs, which eventually fuse to form myotube [ 162 ]. Additionally, miR-139-5p regulates the skeletal myogenic differentiation of human DPSCs by interacting with the Wnt/ β -catenin signaling pathway [ 79 ]. These outcomes reveal that miRNAs are essential for the induction of myogenic differentiation of DPSCs.…”

“…Bone marrow was the first identified and, historically, most frequently utilized source of MSCs. More recently, MSCs have been identified in and isolated from umbilical cord, adipose tissue, molar cells, urine, amniotic fluid, and connective tissue (Dong et al, 2018; Xie and Shen, 2018).…”
