MicroRNA‑182 promotes epithelial‑mesenchymal transition by targeting FOXN3 in gallbladder cancer

Abstract: Increasing evidence has suggested an association between the expression profiles of microRNAs (miRs) and gallbladder cancer (GBC). Recently, miR-182 has been demonstrated to exert tumor-promoting effects. However, the biological activity and molecular mechanisms of miR-182 in GBC remain unclear. The results of the present study demonstrated that miR-182 expression was significantly upregulated in GBC tissues and cell lines (GBC-SD and SGC-996). In addition, miR-182-knockdown attenuated epithelial-mesenchymal t… Show more

“…In diffuse large B-cell lymphoma, lnc SMAD5-AS1 upregulate APCs expression and inhibits cancer cell proliferation as an endogenous competitor of miR-135b-5p, in addition, it inhibits the cancer cell proliferation via Wnt/β-catenin signaling pathway, 18 FOXN3 can mediate tumor cell proliferation and invasion through the Wnt/β-catenin signaling pathway in melanoma, 43 thyroid cancer, 19 and gallbladder cancer. 44 In our study, we measured the expression of related proteins of the Wnt/β-catenin signaling, like β-catenin, Cyclin D1 and C-myc. Moreover, the expression of β-catenin, Cyclin D1 and C-myc was regulated by miR-135b-5p.…”

“…However, additional miR-216a/b targets also may contribute to the phenotypes observed after ectopic expression of miR-216a/b. In addition, regulation of Foxn3 by other miRNAs has been described in non-neural cells, including miR-182 (Zhang et al, 2021), a miRNA highly expressed in retina (Xu et al, 2007;Zhuang et al, 2020), and miR-135b (Han et al, 2019), via a site near miR-216a/b site 1 in the Foxn3 3′ UTR. miR-135b-5p is expressed in retina (Table S2), so combinatorial or redundant miRNA regulation of retinal Foxn3 is possible.…”

“…A549 or A549/DDP cells that achieved 60% confluency (1 × 10 5 cells/well) was applied to transfection by lipofectamine 2000 (Invitrogen), and then cultivated in a day using 6-well plates. The concentrations of miR-1297 mimics as well as miR-NC in the eventual transfection system were set at 50 nM [ 26 ].…”

“…In addition, we constructed pcDNA3.1-p300 vector (RiboBio) through the clone of p300ʹs open reading frame (ORF) in pcDNA3.1 vector. Afterward, we used lipofectamine 2000 (Invitrogen) to transfect pcDNA3.1-p300 vectors in A549 or A549/DDP cells [ 26 ].…”
