MicroRNA-339 and microRNA-556 regulate Klotho expression in vitro

Abstract: Klotho is an anti-aging protein with direct effects on life-span in mice. Klotho functions to regulate pathways classically associated with longevity including insulin/IGF1 and Wnt signaling. Decreased Klotho protein expression is observed throughout the body during the normal aging process. While increased methylation of the Klotho promoter is reported, other epigenetic mechanisms could contribute to age-related downregulation of Klotho expression, including microRNA-mediated regulation. Following in silico i… Show more

“…miRNAs are involved in 3′ UTR translational regulation through binding mRNA to a 3′ UTR region of a gene to repress translation or promote mRNA degradation. The repression of translation in aging and longevity‐related networks has previously been implicated in the aging process (Chen et al ., ; Mehi et al ., ). It is possible that the duplication of genes involved in these processes could impact the evolution of longevity or of other life history traits.…”

“…MicroRNA may also epigenetically regulate Klotho expression. Thus microRNA-339 and microRNA-556 directly decrease Klotho protein expression (65).…”

“…TNF and TNF-like weak inducer of apoptosis-downregulated renal Klotho expression can be blunted by inhibition of histone deacetylase [84] , indicating that cytokine-induced elevation of histone acetylation is associated with downregulation of Klotho expression. Other epigenetic mechanisms such as microRNA-mediated regulation also contribute to the downregulation of Klotho expression in diabetic nephropathy and CKD [85,86] . However, the therapeutic efficiency of epigenetic modulation in the upregulation of renal Klotho is worth confirming in kidney diseases.…”
