MicroRNA expression profile is altered in the upper airway skeletal muscle tissue of patients with obstructive sleep apnea-hypopnea syndrome

Abstract: Objective To investigate the involvement of microRNAs (miRNAs) in the pathogenesis of obstructive sleep apnea-hypopnea syndrome (OSAHS). Methods In this study, we investigated miRNA profiles in the upper airway (UA) skeletal muscles of four patients with OSAHS and four matched controls using the miRCURY miRNA array. In another cohort of 12 OSAHS cases and 7 controls, the mRNA expression levels of interleukin (IL)-6 and Lin-28 homolog A (Lin28A), targets of the downregulated let-7 family members, were measured … Show more

“…miR-101-3p was also downregulated and was previously reported to exhibit similar pattern in neuromyelitis optica (52) and familial dysautonomia patients (53). Moreover, miR-101-3p was also found to be downregulated in the upper airway skeletal muscles of patients suffering from obstructive sleep apnea-hypopnea syndrome (54).…”

“…Based on the normalization counts, the significantly differentially expressed miRNAs in three stages were determined by their fold change. Based on a power analysis using the IDEG6 ( Romualdi et al, 2003 ), we determined that our design had over 99% power to detect differentially expressed miRNAs (DEMs) at | log2(FC) | > = 1 and FDR < = 0.01 ( Hou et al, 2019 ). We then screened for DEMs in the 30 d vs. 60 d and 60 d vs. 90 d pairwise comparisons.…”

“…In addition, a study of the miRNA profiles of OSA in samples of upper airway skeletal muscle was conducted [43]. From the 370 miRNAs identified in the screening phase, hsa-let-7 family miRNAs, hsa-miR-34a-5p, hsa-miR-92a-3p, and hsa-miR-101-3p were downregulated in OSA patients.…”
