MicroRNAs and Adrenocortical Tumors: Where do we Stand on Primary Aldosteronism?

Abstract: MicroRNAs, the endogenous mediators of RNA interference, interact with the renin-angiotensin-aldosterone system, regulate aldosterone secretion and aldosterone effects. Some novel data show that the expression of some microRNAs is altered in primary aldosteronism, and some of these appear to have pathogenic relevance, as well. Differences in the circulating microRNA expression profiles between the two major form… Show more

“…Steroid profiling by liquid chromatograph-mass spectrometry [ 58 ] and detection of increased 18-oxocortisol and 18-hydroxycortisol levels specifically in APAs harboring potassium inwardly rectifying channel subfamily J member 5 ( KCNJ5 ) mutations [ 59 , 60 ] have been proposed as alternative methods to AVS for classifying the PA subtype. In addition, several microRNAs with differential expression in APA tissues and circulating blood [ 61 , 62 ] have been demonstrated as potential biomarkers useful for subtype diagnosis. Further studies to prove the specificity, sensitivity, and cost effectiveness are needed.…”

“…However, the influence of miRNA on the aldosterone steroidogenic pathway is not confined to CYP11B2 ; we have also shown significant effects on CYP11A1 and CYP21A2 [ 19 ], and it would seem likely that miRNAs are able to control this specific pathway at various stages, even before one considers their influence over the various regulatory mechanisms (primarily the renin–angiotensin system) that stimulate the adrenal cortex to produce aldosterone. Therefore, it seems likely that we have only scratched the surface of the numerous ways in which miRNAs intervene in the regulation, synthesis and action of aldosterone, either in the adrenal cortex or elsewhere [ 22 , 23 , 24 ].…”

“…As a potential marker, miRNAs have also been studied in cardiovascular diseases, endocrine diseases, tumors, etc. (30)(31)(32). Depression is one of the most common mental diseases in the clinic (33).…”
