Microsatellite markers for the endangered fir Abies guatemalensis (Pinaceae)

Abstract: Eight newly developed polymorphic microsatellite markers are presented for Abies guatemalensis Rehder, an endangered tree endemic to Mexico and Central America. These microsatellite markers are useful for evaluating the population genetics of the species which is threatened by greenery poaching in natural populations. Novel conservation plans are needed to secure a more sustainable use strategy and to preserve the genetic diversity of the species.

“…DNA extraction protocol according to Doyle and Doyle () was used to extract DNA from approximately 50 mg of silica gel dried needles. Eleven nuclear microsatellite loci previously identified in various Abies species were used SF324, SF333, SFb04, SF1 (Cremer et al., ), ABF18 (Saito, Lian, Hogetsu, & Ide, ), NFF7, NFF3, NFH15, NFH3 (Hansen, Vendramin, Sebastiani, & Edwards, ), AB15 (Rasmussen, Andersen, Frauenfelder, & Kollmann, ), and AfSI16 (Josserand et al., ). Two multiplex and three singleplex reactions were done.…”

“…In the present study, the high level of polymorphism and heterozygosity of 22 microsatellite loci would be a valuable to investigate further population studies with more extensive samplings from A. koreana and / or A. nephrolepis. The values of H E in A. koreana and A. nephrolepis were moderate in the Abies genus (Saito et al, 2005;Cremer et al, 2006;Josserand et al, 2006;Lian et al, 2007;Rasmussen et al, 2008;Sánchez-Robles et al, 2012;Zhan et al, 2014).…”

“…The same procedure was used for further genotyping after the testing step, though here only two PCR reactions were made for each individual: PCR-A containing seven primer pairs: NFH15, NFH3 and NFF3 (Hansen et al 2005a), Ab15 and Ab12 (Rasmussen et al 2008), SF 78 (Cremer et al 2006) and Abfi18 (Saito et al 2005); PCR-B containing six primer pairs: NFH3, NFF2 and NFF7 (Hansen et al 2005a), SF b4 (Cremer et al 2006), As09 (Lian et al 2007) and AfSI_16 (Josserand et al 2006). NFH3 was included in both PCRs as an internal control.…”
