MIF Variant rs755622 Is Associated with Severe Crohn’s Disease and Better Response to Anti-TNF Adalimumab Therapy

Abstract: Crohn’s disease (CD), rheumatoid arthritis, psoriatic arthritis and other inflammatory diseases comprise a group of chronic diseases with immune-mediated pathogenesis which share common pathological pathways, as well as treatment strategies including anti-TNF biologic therapy. However, the response rate to anti-TNF therapy among those diseases varies, and approximately one third of patients do not respond. Since pharmacogenetic studies for anti-TNF therapy have been more frequent for other related diseases and… Show more

“…Patients with the GG allele were shown to have greater disease activity. Similar observations have been made in patients with rheumatoid arthritis and psoriatic arthritis [41]. A new direction of research and the role of genetics in response to biological therapy are bioinformatics analyses of many cohorts [17].…”

“…Thus, SNPs in this gene could affect the bacterial composition of the gut of patients with IBD and immune recognition. In this context, the presence of rs10210302 correlates in CD with a beneficial response to ADA therapy [61,62]. SNP rs1295686, in the IL-13 gene, is of biological interest given that it is implicated, as a functional enhancer, with the TH2LCRR promoter (T helper type 2 locus control region-associated RNA).…”
