Milk lymphocyte profile and macrophage functions: new insights into the immunity of the mammary gland in quarters infected with Corynebacterium bovis

Abstract: Backgrounds The present study explored the viability of bovine milk macrophages, their intracellular production of reactive oxygen and nitrogen species (RONS), and their phagocytosis of Staphylococcus aureus, as well as the profile of lymphocytes, from healthy udder quarters and udder quarters infected by Corynebacterium bovis. The study included 28 healthy udder quarters from 12 dairy cows and 20 udder quarters infected by C. bovis from 10 dairy cows. The percentages of macrophages and lymphoc… Show more

“…The impact of TLR priming on the phagocytosis activity of purified neutrophils was analyzed using flow cytometry. [ 24 ]. For this, heat-killed S. aureus (Pansorbin) was purchased from Calbiochem (Nottingham, UK) and labeled with fluoresceinisothiocyanate (FITC) according to the manufacturer’s protocol (fluoresceinisothiocyanate labeling kit, Sigma-Aldrich, St. Louis, MO, USA).…”

“…Bacterial phagocytosis by milk phagocytes was performed using heat-killed staphylococcus aureus ( S. aureus ) bacteria (Pansorbin, Calbiochem, Merck, Nottingham, UK) labeled with fluoresceinisothiocyanate (FITC, Sigma-Aldrich, St. Louis, Missouri, USA) ( 54 ). Separated milk cells (2 × 10 5 in 100 μl RPMI medium) were incubated with S. aureus-FITC (50 bacteria/cell) for 45 min at 37°C in 96 well plates.…”

“…The bacterial phagocytosis by camel MDM was performed by incubating MDM with heat-killed Staphylococcus aureus (S. aureus) bacteria (Calbiochem, Nottingham, UK) labeled with a FITC labeling kit (Sigma-Aldrich, Missouri, USA) according to the manufacturer instructions (Silva et al, 2021). Camel MDM (1 x 10 3 cells in 100 µL RPMI medium) were incubated with S. aureus-FITC (20 bacteria/cell) for 30 minutes at 37°C and 5% CO2.…”
