Mimicking of Estradiol Binding by Flame Retardants and Their Metabolites: A Crystallographic Analysis

Abstract: Background: Brominated flame retardants (BFRs), used in many types of consumer goods, are being studied because of concerns about possible health effects related to endocrine disruption, immunotoxicity, reproductive toxicity, and neurotoxicity. Tetrabromobisphenol A (TBBPA), the most widely used BFR, and human metabolites of certain congeners of polybrominated diphenyl ether (e.g., 3-OH-BDE-47) have been suggested to inhibit estrogen sulfotransferase, potentially affecting estrogen metabolism. Objectives: Our p… Show more

“…Binding distance has been used as a tool for docking scoring and for evaluation of contact between ligand and LBD (Baker and Chandsawangbhuwana, 2012;Gosavi et al, 2013;Park et al, 2011). Alteration of binding distance between ligand and each binding domain accounts for changes in estrogenic potency of 4-methyl-2,4-bis(4-hydrozyphenyl)pent-1-ene (MBP), a bisphenol A (BPA) metabolite, compared to that of its unsubstituted compound (Baker and Chandsawangbhuwana, 2012).…”

“…The same group also reported hydroxylated BDEs to have mutagenic potential and exhibit dioxin-like toxicity as AhR-agonists (Su et al, 2012a(Su et al, , 2014. In addition, OH-BDEs were also observed to inhibit the enzyme SULT1E1 in humans and potentially affect estrogen metabolism (Gosavi et al, 2013).…”

“…Brominated flame retardants, PBDEs, are an example of EDCs that are both persistent and bio accumulative; they can mimic the female hormone oestrogen and thus interfere with the action of the body's natural hormones. (Gosavi RA, et al, 2013).…”
