Minicells, Back in Fashion

Abstract: bCryo-electron tomography (cryo-ET) has emerged as a leading technique for three-dimensional visualization of large macromolecular complexes and their conformational changes in their native cellular environment. However, the resolution and potential applications of cryo-ET are fundamentally limited by specimen thickness, preventing high-resolution in situ visualization of macromolecular structures in many bacteria (such as Escherichia coli and Salmonella enterica). Minicells, which were discovered nearly 50 ye… Show more

“…This extreme situation (CV 2 p = 1) appears, however, only when one daughter inherits all of the mother's size resulting in a non-growing cell and one that grows without bound. Such a situation has been observed in minicell-producing mutants of E. coli, but these cells also allow for divisions at mid-cell reinforcing their overall viability [33]. In the following, we investigate in more detail the conditions for finite mean and variance.…”

“…Under these circumstances, FtsZ will promote asymmetric cell division, thus resulting in the formation of minicells, which usually do not contain nucleic acids and are thus unable to further divide . Historically, minicells have been exploited as model systems for visualizing molecular machines by using high‐resolution cryoelectron tomography . As the resolution of cryoelectron tomography is limited by the species thickness, minicells are attractive due to their very small size.…”
