Minocycline enhances the mesenchymal stromal/stem cell pro-healing phenotype in triple antimicrobial-loaded hydrogels

Abstract: Mesenchymal stromal/stem cells (MSCs) have demonstrated pro-healing properties including an anti-inflammatory cytokine profile and the promotion of angiogenesis via expression of growth factors in pre-clinical models. MSCs encapsulated in poly(ethylene glycol) diacrylate (PEGdA) and thiolated gelatin poly(ethylene glycol) (Gel-PEG-Cys) crosslinked hydrogels have led to controlled cellular presentation at wound sites with favorable wound healing outcomes. However, the therapeutic potential of MSC-loaded hydroge… Show more

“…We have shown previously that minocycline can enhance the wound healing properties in human bone marrow MSCs, but the mechanism of enhancement was still unknown [17]. Here, we demonstrate that this enhancement of the wound healing phenotype may be due to the phosphorylation and potential activation of NFκB in MSCs.…”

“…Hydrogel formulations were made in PBS to synthesize a 0.5% (w/v) Irgacure 2959 photoinitiator (BASF, Ludwigshafen, Germany), 10% (w/v) PEGdA and 10% (w/v) Gel-PEG-Cys sterile-filtered solution prepared and characterized as described previously 15 - 16 , 24 . The procedures for the preparation of M1 hydrogels used in vitro and in vivo studies was illustrated ( Figure 1 ).…”

“…Minocycline induces the following favorable changes in MSCs: migratory capacity, proliferation, gene expression, and growth factor release, ultimately resulting in enhancement of angiogenesis. Also, the triple antimicrobial-loaded hydrogels reduce bacterial bioburden and preserve viability of MSCs in the presence of bacteria [71].…”
