miR-10a rejuvenates aged human mesenchymal stem cells and improves heart function after myocardial infarction through KLF4

Abstract: BackgroundAging is one of the key factors that regulate the function of human bone marrow mesenchymal stem cells (hBM-MSCs) and related changes in microRNA (miRNA) expression. However, data reported on aging-related miRNA changes in hBM-MSCs are limited.MethodsWe demonstrated previously that miR-10a is significantly decreased in aged hBM-MSCs and restoration of the miR-10a level attenuated cell senescence and increased the differentiation capacity of aged hBM-MSCs by repressing Krüpple-like factor 4 (KLF4). In… Show more

“…Overexpression of neuron-derived neurotrophic factor was found to rejuvenate aged BM-MSCs and improve their function in repairing the aged heart after ischemia [82]. microRNA (miR)-10a rejuvenated aged BM-MSCs and enhanced the cardiacprotection following infarction in mice via increased paracrine effects [83]. Furthermore, overexpressing FGF 21 in MSCs may delay their senescence during passaging in vitro [84].…”

“…The overexpression of miR-10a in aged hBM-MSCs stimulates angiogenesis by inducing the expression of angiogenic factors via activated Akt. These cells then enhance implanted stem cell survival and improve cardiac function after MI[ 129 ]. The miR-29b-3p derived from BM-MSCs regulates aging-associated insulin resistance[ 132 ].…”

“…Moreover, miR-10a has been suggested as a biomarker for heart failure prognosis, since it is abnormally expressed in rats with heart failure [15]. It can also promote the expression of angiogenic factors to improve angiogenesis and cardiac function in mice after myocardial infarction [16]. Of note, miR-10a can promote the progression of hepatic fibrosis through modulating the TGFβ1/Smads signaling pathway [17].…”
