MiR-1224-5p attenuates polycystic ovary syndrome through inhibiting NOD-like receptor protein 3 inflammasome activation via targeting Forkhead box O 1

Abstract: Polycystic ovary syndrome (PCOS) is a common endocrine disorder that poses a great threat to women's health. MiR-1224-5p is downregulated in the follicular fluid of patients with PCOS, but its role remains largely unknown. In this study, mice were treated with dehydroepiandrosterone (DHEA) to establish an in vivo model of PCOS. We found that enhanced activation of NLRP3 inflammasome was accompanied by downregulation of miR-1224-5p in ovarian tissue of PCOS mice. The effect of miR-1224-5p was further explored i… Show more

“…In PCOS, miR-1224-5p deactivates the NLRP3 inflammasome via FoxO1 [ 130 ]. The ability of granulosa cells to produce mature oocytes is arrested because of inflammatory reactions, which have also been confirmed in patients with PCOS [ 131 ].…”

“…(2021) [104] In vitro: Upregulation of miR-1224-5p − Inhibited the production of testosterone and DHEA − Attenuated inflammation by suppressing NF-κB p65, NLRP3 inflammasome, and IL-1β − Decreased the expression of FOXO1 and its downstream gene TXNIP − FOXO1 was confirmed as a target of miR-1224-5p. − The downregulation of miR-1224-5p was found to be linked to an elevated activation of the NLRP3 inflammasome.…”

“…Another study demonstrated that CCL2 as the main target of miR-374a-5p, and also demonstrated that the transcription factor NF-kB can activate CCL2, suggesting that miR-374a-5p may play a role in regulating inflammatory responses (109). Li et al described that miR-1224-5p exerts anti-inflammatory effects and alleviates PCOS by inhibiting the activation of the NF-kB signaling pathway (141).…”
