MiR-21-5p regulates the dynamic of mitochondria network and rejuvenates the senile phenotype of bone marrow stromal cells (BMSCs) isolated from osteoporotic SAM/P6 mice

Abstract: Background Progression of senile osteoporosis is associated with deteriorated regenerative potential of bone marrow-derived mesenchymal stem/stromal cells (BMSCs). According to the recent results, the senescent phenotype of osteoporotic cells strongly correlates with impaired regulation of mitochondria dynamics. Moreover, due to the ageing of population and growing osteoporosis incidence, more efficient methods concerning BMSCs rejuvenation are intensely investigated. Recently, miR-21-5p was re… Show more

“…In fact, miR-21-5p has been shown to be a positive regulator of osteoblast differentiation and function, suggesting that its knockdown might impair osteoblast function. 49 , 50 Second, even though the mRNA expression of Nrf2 was restored in diabetic bone after antagomir treatment, Keap 1 expression, the negative regulator of Nrf2, tended to be increased suggesting an incomplete activation of Nrf2 in response to the antagomir treatment. This may go in line with the unchanged levels of Hmox1, one of the main targeted genes of Nrf2 with antioxidant and cytoprotective effects.…”

“…MiR-21 also plays a crucial role in promoting bone formation [ 34 ]. Previous studies have demonstrated that transfection of miR-21-5p mimics into MC3T3-E1 cells enhanced the expression of Runx2 and Osterix through the miR-21-5p/Dec1 axis [ 35 ].…”

“…However, upregulation of miR-21 induces mitochondrial fission, resulting in a decrease in the proportion of tubular mitochondria and an increase in globular mitochondria. Moreover, BMSCs obtained from simple senile osteoporosis patients show down-regulated expression of MFN-1 and MFF (75). Additionally, oxidative stress-induced damage to osteoblasts leads to increased mitochondrial fission that subsequently impairs both mitochondrial morphology and cellular function.…”
