MiR-25-3p regulates the differentiation of intramuscular preadipocytes in goat via targeting KLF4

Abstract: Abstract. Adipocyte differentiation, which plays an important role in fat deposition, involves a complex molecular mechanism. MicroRNAs (miRNAs) are essential in this progress. Here, we showed that miR-25-3p expression had increased during goat intramuscular preadipocyte differentiation, which peaked at day 3. Using liposome transfection and qRT-PCR techniques, we found that knocking down miR-25-3p reduced the accumulation of lipid droplets by downregulating or upregulating the expression of LPL, PPARγ, AP2, S… Show more

“…Goat intramuscular preadipocytes (GIMPA) were isolated from the longissimus dorsi muscle (Sichuan jianyang dageda animal husbandry CO., LTD, Sichuan, China) of 7-day-old Jianzhou Da-er goats. The isolation and culture of cells was consistent with the previous study [ 25 ]. DMEM-F12 (Gibco, Carlsbad, CA, USA) containing 10% FBS (fetal bovine serum, Gibco, USA), 1‰ Penicillin-Streptomycin (Gibco, Carlsbad, CA, USA) and 50 μmol·L −1 oleic acid (Sigma, St Louis, MO, USA) was used to induce adipogenic differentiation of GIMPA to goat intramuscular adipocytes (GIMA).…”

“…ncRNAs, including microRNAs (miRNAs), long non-coding RNAs (lncRNAs) and circular RNAs (circRNAs), have also been shown to be important regulators of adipocyte differentiation in recent years [ 21 , 22 , 23 ]. For instance, miR-340-5p inhibits ovine adipocyte differentiation by targeting ATF7 [ 24 ] and miR-25-3p regulates the differentiation of intramuscular preadipocytes in goats via targeting KLF4 [ 25 ]. He et al (2022) previously constructed an lncRNA-miRNA-mRNA regulatory network during goat intramuscular and subcutaneous adipocyte differentiation by RNA-seq, and identified several lncRNAs that may regulate adipocyte differentiation [ 26 ].…”

“…Currently, thousands of miRNA gene codes have been recognized [ 37 ]. The interaction involves the formation of a double-stranded assembly between the miRNA seed and the mRNA “target” [ 38 , 39 ].…”

“…Currently, thousands of miRNA gene codes have been recognized [37]. The interaction involves the formation of a double-stranded assembly between the miRNA seed and the mRNA "target" [38,39]. Kim et al 2010 [40] have described the relationship between miRNAs and the regulation of early adipocyte differentiation.…”
