miR-29b-3p Inhibitor Alleviates Hypomethylation-Related Aberrations Through a Feedback Loop Between miR-29b-3p and DNA Methylation in Cardiomyocytes

Abstract: As a member of the miR-29 family, miR-29b regulates global DNA methylation through target DNA methyltransferases (DNMTs) and acts as both a target and a key effector in DNA methylation. In this study, we found that miR-29b-3p expression was inversely correlated with DNMT expression in the heart tissues of patients with congenital heart disease (CHD), but whether it interacts with DNMTs in cardiomyocytes remains unknown. Further results revealed a feedback loop between miR-29b-3p and DNMTs in cardiomyocytes. Mo… Show more

“…MiR-15a/miR-16-1 can interact with HDAC3 and HDAC9, which affects histone activity and the expression of cell cycle-related genes [346,348]. However, miR-29b interacts with DNMT3A and DNMT3B, affecting DNA methylation and the expression of key genes used in the cell cycle [317,346,349]. In colorectal cancer, miRNAs such as miR-449a have been implicated in cell cycle regulation through their interactions with histone-modifying enzymes.…”

“…For instance, DNA methyltransferases and the miR-29b-3p levels were negatively correlated in the CHD patients [ 238 ]. For cardiac cell proliferation and cardiac abnormalities, certain miRNAs (hsa-miR-221-3p, hsa-miR-218-5p, and hsa-miR-873-5p) have been described in [ 239 ].…”
