MiR-337-3p Promotes Adipocyte Browning by Inhibiting TWIST1

Abstract: The prevalence of metabolic syndrome (MetS) and obesity is an alarming health issue worldwide. Obesity is characterized by an excessive accumulation of white adipose tissue (WAT), and it is associated with diminished brown adipose tissue (BAT) activity. Twist1 acts as a negative feedback regulator of BAT metabolism. Therefore, targeting Twist1 could become a strategy for obesity and metabolic disease. Here, we have identified miR-337-3p as an upstream regulator of Twist1. Increased miR-337-3p expression parall… Show more

“…MicroRNAs (miRNAs, miRs) are small, highly conserved, non-coding single-stranded ribonucleic acids (RNAs), which enhance messenger RNA (mRNA) degradation and/or inhibit protein translation by binding to the 3 -untranslated regions (UTRs) of target mRNAs [31][32][33][34]. They play essential regulatory roles in a number of biological processes, both in health and in disease, and have been extensively investigated in cardiovascular medicine [35][36][37][38].…”

“…Let-7 is one of the well-studied miRNAs which inhibits pre-adipocytes clonal expansion and terminal differentiation via targeting HMGA2 (Sun et al, 2009). MiR-337-3p can inhibit TWIST1 and promote transcription of genes participating in brown fat metabolism (Vonhögen et al, 2020). In circ_0000819, two miR-337-3p binding sites were identified.…”

“…The expression level of miR-337-3p in the BAT was significantly higher than that in the WAT. Also, the overexpression of miR-337-3p in brown preadipocytes resulted in the increased expression of mitochondrial markers, thus suggesting that miR-337-3p can enhance adipocyte browning [77]. MiR-30a is enriched in the BAT and the forced expression of miR-30a increases mitochondrial volume and mitochondrial respiration in human white adipocytes, making them characteristic of brown adipocytes [98].…”
