miR-663 Suppresses Oncogenic Function of CXCR4 in Glioblastoma

Abstract: Purpose: To identify the miRNA regulators of C-X-C motif chemokine receptor 4 (CXCR4) and the underlying mechanism as well as the therapeutic and prognostic values in human glioblastoma (GBM).Experimental Design: miRNA profile analyses and bioinformatics predictions were used to identify the mediators of CXCR4, which were confirmed by luciferase reporter assay, Western blot assay and immunohistochemistry. The effects of miR-663 on CXCR4-mediated GBM malignancy were investigated by gainof-function experiments. … Show more

“…The present study shows that CXCR4 expression was reduced in over-expressing miR-663a HCT116 cells, which resulted in the suppression of proliferation. Yu et al reports that miR-663a negatively regulates CXCR4 expression by targeting its coding sequence in human glioblastoma cells and compromises proliferation [34]. This report suggests that miR-663a directly binds to the coding sequence of CXCR4 mRNA to suppress its translation.…”

“…qRT-PCR was performed using a Bio-Rad CFX96 Real-Time PCR Detection System (Bio-rad, Hercules, CA, USA) or an ABI 7900HT Sequence Detection System (Applied Biosystems, Foster City, CA, USA) as previously described. 8,44 The primer sequences were listed in Supplementary Table S6. Expression of GAPDH was used for the normalization.…”

“…CXCR4 and NR5A2 are all reported to be associated with the progression of human cancers [49][50][51][52]. Besides, CXCR4 has been proved to be related to GBM [53,54]. As to NR5A2, it encodes a DNA-binding zinc finger transcription factor protein and involved in the expression of genes for hepatitis B virus and cholesterol biosynthesis.…”
