miRNA-10b sponge: An anti-breast cancer study in vitro

Abstract: Breast cancer is a malignant tumor with the highest incidence among women. Breast cancer metastasis is the major cause of treatment failure and mortality among such patients. MicroRNAs (miRNAs) are a class of small molecular non-coding regulatory RNAs, which act as oncogenes or tumor suppressors in breast cancer. miRNA-10b has been found to exhibit a high expression level in advanced and metastatic breast cancer, and is closely related to breast cancer metastasis. An miRNA sponge is an mRNA with several repeat… Show more

“…Currently, apoptotic resistance is an important factor in chemotherapy-managed metastatic ovarian cancer in patients (7,8). The biological relevance of Chinese medicinal therapy in the progression of breast cancer is evidenced by the increasing immunosuppression, which has demonstrated their capacities to induce apoptosis and inhibit tumor aggressiveness (9,10). Therefore, the development of Chinese medicine monomers that overcome apoptotic resistance has been the focus of investigations in the treatment of cancer.…”

“…Overexpression of miR-10b in breast cancer cells can inhibit the expression 25 Huang et al, 32 Bahena-Ocampo et al, 33 Liang et al, 34 Ma et al, 35 Petrovic, 36 Yan et al 37 51 Rasheed et al, 52 Tavazoie et al, 53 Chou et al, 54 Zhang et al 55 43 A study showed that the upregulation of miR-155 can facilitate tumor angiogenesis, while it is associated with a poor prognosis for triple-negative breast cancer patients. 44 Kong et al 45 reported that miR-155 can act on TGF-β/Smad4 signaling through RhoA, thereby promoting the EMT; in contrast, knockdown of miR-155 can significantly inhibit the TGF-β/Smad4-induced EMT.…”

“…For example, miRNA sponges designed for modulation of miR-10b, miR-21, miR-155 and miR-221/222 mediate an impact on the activity of many protein targets, such as HOXD-10, PDCD4, Smad4, SRC3, Bcl-2 Bim, FOXO3a, PTEN and RhoA. Thus they provide inhibition of proliferation, activation of apoptosis and increase in the sensitivity of tumour cells to chemotherapy [28][29][30]. Suppression of oncogenic activity of miR-522 in a non-small cell lung M A N U S C R I P T…”
