Summary. Non-esterified fatty acid (NEFA) contents were determined (1) in cow, goat, mare and human plasmas or sera and (2) in bovine serum albumin solutions by thin-layer and gas-liquid chromatographic methods which were compared with 6 chemical methods of determination and 2 enzymatic methods (manual and automatic). The chemical methods combined two extraction techniques (isopropanol-heptane and silicic acid-diisopropyl ether) and three determination methods (titrimetry and colorimetry using either phenol red or copper soaps). Correlations between the nine various methods were high, but in some cases regression lines were different from the bissector and species-dependent. The putative occurrence of interfering compounds or the possible effect of between-species differences in NEFA composition have been discussed. The choice of a fast method depends primarily on practical considerations, but the need to standardize each method for a given species has been stressed. Automatic enzymatic assay seemed to be suitable for routine analyses.Introduction.

-The influence of maternal and formula milk on lipid metabolism was studied in 7-day-old pigs. Lipid content, fatty acid composition, lipogenic enzyme activities and expression of GLUT4 mRNA were determined in subcutaneous adipose tissue and skeletal muscle from pigs that were bottle-fed formula milk (F) or sow milk (SM), or were sow-reared (SR). Bottle-fed pigs were isoenergetically fed and achieved similar daily body weight gain. SR pigs have a higher (P < 0.05) body weight gain than bottle-fed pigs. Lipid content of adipose tissue was lower (P < 0.05) in F than in SM and SR pigs. In muscle, lipid content did not differ significantly between groups. In adipose tissue, acetyl-CoA-carboxylase (CBX), fatty acid synthase (FAS), malic enzyme (ME), glucose-6-phosphate-dehydrogenase (G6PDH) and lipoprotein lipase (LPL) activities and GLUT4 mRNA levels were higher (P < 0.05) in SR than in bottle-fed pigs. In muscle, ME and G6PDH activities and GLUT4 mRNA were higher (P < 0.05) in F than in SM and SR pigs; LPL was not detected. It has been shown that the same modifications on the composition of brown adipose tissue (BAT) which are normally induced following cold stimulation are also observed in hypophysectomized rats acclimated either at 28 degrees C or 15 degrees C. To test the possibility of BAT stimulation in hypophysectomized rats, we have determined some enzymatic activities known to modulate the energy supply to that organ. Seven week old Long-Evans rats were hypophysectomized. Three weeks later, they were exposed to either 28 degrees C or 15 degrees C ambient temperature for five or six weeks. Hypophysectomized rats were compared to age matched or weight matched controls. Total lipoprotein lipase activity (LPL) (triglyceride uptake) was enhanced in BAT of 28 degrees C hypophysectomized rats compared to controls. Cold acclimation led to a large increased activity. Total LPL activity was comparable in BAT of hypophysectomized and control rats. Total malic enzyme and glucose-6-phosphate dehydrogenase activities (in situ lipogenesis) were doubled in BAT of 28 degrees C hypophysectomized compared to controls. A large enhancement was observed in BAT of either 15 degrees C control or 15 degrees C hypophysectomized rats. Among the studied organs (liver, white adipose tissue, heart, BAT) hypophysectomy promotes the three enzyme activities only in BAT. These variations were discussed with relation to the effect of hypophysectomy on brown adipose tissue at 15 degrees C and 28 degrees C.
