Mitochondrial Contact Site and Cristae Organization System and F 1 F O -ATP Synthase Crosstalk Is a Fundamental Property of Mitochondrial Cristae

Abstract: Mitochondria have undergone profound diversification in separate lineages that have radiated since the last common ancestor of eukaryotes some eons ago. Most eukaryotes are unicellular protists, including etiological agents of infectious diseases, like Trypanosoma brucei .

“…Interestingly, Tb927.8.3070 has not been detected in previous pull-down MS analyses of the T. brucei F 1 F o complexes ( 33 , 61 ). This could be explained by complex disruption during extraction and/or by its low molecular mass and having few tryptic cleavage sites, which may make detection by proteomic methods difficult ( 62 ).…”

“…However, should this be the case, no similarity with subunit b from Euglena or from other organisms would be expected. Interestingly, Tb927.8.3070 has not been detected in previous pull-down MS analyses of the T. brucei F1Fo complexes (33,61). This could be explained by complex disruption during extraction and/or by the low molecular mass and the few tryptic cleavage sites of subunit b which may make detection by proteomic methods difficult (62).…”

“…The remaining beads from triplicate APs of Mic60-6xHis, Orf52-6xHis and the mock controls were trypsinized and analyzed by LC-MS/MS. Protein enrichment in bait-APs in comparison to the mock control was quantified using label-free quantification as previously described (Cadena et al 2021). In each AP, 497 high-confidence proteins were found (Dataset 2) based on mean Andromeda confidence scores >100 in three biological replicates (Cox et al 2011).…”
