Mitochondrial D-loop diversity of grasscutter (<i>Thryonomys swinderianus</i> Rodentia: Hystricomorpha) in Ghana

Abstract: Attempts are being made to domesticate the grasscutter (Thryonomys swinderianus) for commercial production in Sub-Saharan Africa to cater for the protein needs of the people and to satisfy the craving for bushmeat, thereby reducing habitat destruction through hunting. The objective of this study was to determine the genetic diversity of grasscutter populations in Ghana. DNA was extracted from roots of hair samples collected from 84 grasscutters from three agro-ecological zones in Ghana, namely Guinea Savanna (… Show more

“…Other works reported in the literature include: characterization of the estrus cycle, determination of the ovulatory mechanisms, detection of mating and pregnancy, efficacy of vaginal mucous plug formation in pregnancy detection, and efficacy of sex determination using ano‐genital distance, amongst others. More recently, advanced techniques such as the use of mitochondrial D‐loop and double‐digest restriction‐site associated DNA (RAD) sequencing markers (single nucleotide polymorphisms, SNPs) have been deployed to improve understanding of genetic diversity in the GCR and for genetic management of production …”

“…Amplification of a ~650 bp fragment of the cytochrome c oxidase I (COI) gene was performed using primers designed by Gaubert et al (2014). A fragment of ~500 bp was amplified for the mitochondrial D-loop region using primers reported by Adenyo et al (2013). PCR setup was performed in a DNA free laboratory, and all working surfaces were decontaminated using a 1.25% hypochlorite solution, prior to setup.…”

“…Previously reported COI (n = 23; accession numbers: KJ192912-KJ192913, Equatorial Guinea; KJ192933-KJ192949, Ghana; KJ192950-KJ192953 and KJ192955, Nigeria) and D-loop (n = 26; accession numbers: AB675385-AB675410, Ghana) sequences were downloaded from GenBank for downstream analyses (Adenyo et al 2013;Gaubert et al 2014). Sequence assembly and editing of sequences obtained from the current study were performed with GENEIOUS R9 software (Kearse et al 2012).…”

“…Previous studies on grasscutter at the population genetics level are scarce and with limited scope. For example, Adenyo et al (2013) studied exclusively Ghanaian grasscutter population using mitochondrial D-loop region, while other studies have employed microsatellite markers (Adenyo et al, 2017;Coker et al, 2017). Another study on bush meat included grasscutter mitochondrial markers (Gaubert et al, 2015) but did not focus on grasscutter population genetics.…”
