Mitochondrial Damage Associated Molecular Patterns From Femoral Reamings Activate Neutrophils Through Formyl Peptide Receptors and P44/42 MAP Kinase

Abstract: Hypothesis-Fractures and femoral reaming are associated with lung injury. The mechanisms linking fractures and inflammation are unclear; but tissue disruption might release mitochondria. Mitochondria are evolutionarily derived from bacteria and contain "Damage Associated Molecular Patterns" (DAMPs) like formylated peptides that can activate immunocytes. We therefore studied whether fracture reaming releases mitochondrial DAMPs (MTD) and how MTD act on immune cells.Methods-Femur fracture reamings (FFx) from 10 … Show more

“…FPR1 is activated by Nformyl peptides, which are derived from either bacterial peptides or mitochondrial proteins (52,53). The endogenous damageassociated molecular patterns from bone and liver mitochondria can activate neutrophils through FPR1 and induce severe inflammatory response syndrome (54)(55)(56). Therefore, concerns have been raised about the potential of functional FPR1 as a therapeutic target for the development of new drugs to treat inflammatory diseases (21,57).…”

“…ERK1/2 and p38 MAPK signaling is crucial for FAM3D-induced neutrophil activation Activation of neutrophils through formyl peptide receptors depends on MAPK signaling (Hauser et al, 2010). However, whether the activation of the MAPKs is required for FAM3D-induced neutrophil activation and function has not been determined.…”
