Mitochondrial-derived peptides in energy metabolism

Abstract: Mitochondrial-derived peptides (MDPs) are small bioactive peptides encoded by short open reading frames (sORF) in mitochondrial DNA that do not necessarily have traditional hallmarks of protein-coding genes. To date, eight MDPs have been identified, all of which have been shown to have various cyto- or metabolo-protective properties. The 12S ribosomal RNA (MT-RNR1) gene harbors the sequence for MOTS-c, while the other seven MDPs, [humanin and small humanin-like peptides (SHLP) 1-6] are encoded by the 16S ribos… Show more

“…Such regulatory age-determined signaling from mitochondrion to nucleus, lying at the nexus of metabolic regulation, has been neatly demonstrated for replicative aging in yeast [ 48 ]. Interestingly, an important role in retrograde signaling was ascribed to mitochondria-generated peptides [ 49 ].…”

“…Our ndings are consistent with previous studies that have identi ed a relationship between MDPs and insulin resistance. There is evidence of the insulin-sensitizing properties of MDPs mainly derived from cellular and animal models [11]. MOTSc is detected in the circulation, and its target organs are primarily skeletal muscle and fat.…”

“…Localization of many coding and non-coding RNA, components of these granules, demonstrates complex dynamics [ 187 , 188 ]. It is curious that the granules are also associated with the mitochondrial ribosomal RNA, which is found at this moment in the cytoplasm [ 189 , 190 ] and can be translated [ 191 ]. The complex pattern and dynamics of mRNA distribution seem to provide regulation of translation during the maturation of germ cells and in the early stages of embryogenesis [ 185 ].…”
