Mitochondrial DNMT3A and DNA methylation in skeletal muscle and CNS of transgenic mouse models of ALS

Abstract: Cytosine methylation is an epigenetic modification of DNA catalyzed by DNA methyltransferases. Cytosine methylation of mitochondrial DNA (mtDNA) is believed to have relative underrepresentation; however, possible tissue and cell differences in mtDNA methylation and relationships to neurodegenerative disease have not been examined. We show by immunoblotting that DNA methyltransferase 3A (Dnmt3a) isoform is present in pure mitochondria of adult mouse CNS, skeletal muscle, and testes, and adult human cerebral cor… Show more

“…Putting the pieces together, evidences largely suggest the presence of methyl cytosines in the mitochondrial DNA, albeit at low levels (Iacobazzi et al, 2013;Wong et al, 2013). This is also supported by the evidence suggesting the localisation of DNA methyltransferases and the TET genes in the mitochondria (Shock et al, 2011).…”

“…Wong and coworkers characterized DNA methylation across two loci in the mitochondrial genome (Wong et al, 2013). The authors suggested that the DNA methyltransferase Dnmt3a is localized to the mitochondria in a tissue specific manner.…”

“…Chestnut and co-workers (2011) also demonstrated that overexpression of Dnmt3a induces degeneration of neuronal NSC34 cells. On the other hand, mitochondrial Dnmt3a protein level is reduced in the skeletal muscle and spinal cord of transgenic SOD1 mouse models of ALS (Wong et al, 2013). CgB has also previously been suggested to play a role in neurodegenerative diseases.…”

“…There is some evidence suggesting that environmental exposures, such as smoking and exposure to pesticides, metals, and other chemicals, may be risk factors [90]. Mitochondria have been implicated in the biological mechanism of the disease, and altered mitochondrial Dnmt3a protein levels and 16S rRNA gene methylation have been observed in mouse models of ALS [91].…”
