Mitochondrial function, morphology and metabolic parameters improve after switching from stavudine to a tenofovir-containing regimen

Abstract: The results from this study demonstrate that systemic and peripheral fat mitochondria improve in patients switched to tenofovir following long-term exposure to stavudine, while continuing protease inhibitor therapy.

“…Moreover, functional alterations in these organelles are typically seen in metabolic diseases related to lipids such as obesity and type 2 diabetes (11,12). In- Age-related lipid metabolism in lipodystrophy J Clin Endocrinol Metab terestingly, the loss of inner-membrane cristae has been also described in human aging cells (24) as well as in adipose tissue of patients with acquired lipodystrophy due to antiretroviral therapy (ART) (25). Also, dilation of ER cisternae is frequently observed in age-related neurodegenerative diseases, and in chronic diseases involving inflammation, as well as in aged animals (26).…”

“…Membranes were developed on chemilumiimager (Roche). Quantitation of the break frequency of mtDNA 8-oxo-dG was based on comparing the undigested and digested mtDNA break frequencies calculated using the Poisson distribution, with units reported in break frequency (BF) (Gerschenson et al, 2009). …”

“…[53][54][55][56] Other studies have reported a lowering of mitochondrial dysfunction when drug regimens were changed and NRTIs were replaced with tenofovir. [57][58][59] A recent study in rats found that very high doses (ie, 100 mg/kg/day) of tenofovir reduced mitochondrial DNA and gene expression in kidney cells. 60 The relevance of this observation given the very high dosing used is unclear.…”
