Mitochondrial Peroxiredoxin 3 Is Rapidly Oxidized and Hyperoxidized by Fatty Acid Hydroperoxides

Abstract: Human peroxiredoxin 3 (HsPrx3) is a thiol-based peroxidase responsible for the reduction of most hydrogen peroxide and peroxynitrite formed in mitochondria. Mitochondrial disfunction can lead to membrane lipoperoxidation, resulting in the formation of lipid-bound fatty acid hydroperoxides (LFA-OOHs) which can be released to become free fatty acid hydroperoxides (fFA-OOHs). Herein, we report that HsPrx3 is oxidized and hyperoxidized by fFA-OOHs including those derived from arachidonic acid and eicosapentaenoic … Show more

“…The balance between the removal and production of H 2 O 2 in mitochondria must be tightly regulated, but our results suggest that a PrxIII defect tips the scales in favor of production in hepatocytes, and to a greater extent in hepatocytes subjected to triggers of apoptosis such as diclofenac. In addition to H 2 O 2 , PrxIII acts on free or lipid-bound fatty acid hydroperoxides [40]. PrxIII is also an efficient mitochondrial peroxynitrite reductase [41].…”

“…Second, hydrogen peroxide, and species with an O-O bond like peroxynitrite and LOOH (Cardozo et al, 2023), react fast with PRDX isoforms (Armas et al, 2019;Bryk et al, 2000;Karplus, 2015). Their reaction with the catalytic cysteine of most PRDX isoforms produces a sulfenic acid, which condenses with a neighbouring cysteine, to form a disulfide crosslink (Peskin et al, 2007;Wood et al, 2003).…”
