Modeling of the enzymatic kinetic synthesis of cephalexin—Influence of substrate concentration and temperature

Abstract: During enzymatic kinetic synthesis of cephalexin, an activated phenylglycine derivative (phenylglycine amide or phenylglycine methyl ester) is coupled to the nucleus 7-aminodeacetoxycephalosporanic acid (7-ADCA). Simultaneously, hydrolysis of phenylglycine amide and hydrolysis of cephalexin take place. This results in a temporary high-product concentration that is subsequently consumed by the enzyme. To optimize productivity, it is necessary to develop models that predict the course of the reaction. Such model… Show more

“…Enzymatic synthesis of Cephalexin involves the combination of 7-aminodeacetylcephalosporanic acid (7-ADCA) and phenylglycine amide in aqueous solution with the enzyme penicillin acylase [2]. Purification of Cephalexin from this dilute reaction mixture is challenging due to the similar structures of Cephalexin and the starting material 7-ADCA [3].…”

“…Among those reactions, the synthesis of semisynthetic penicillins and cephalosporins from the corresponding β-lactam nuclei and suitable acyl donors is of paramount importance to the pharmaceutical industry [7]. Synthesis can be conducted under thermodynamic [8,9] or kinetic [10,11] control. Under thermodynamic control, substrate conversion is limited by the equilibrium of the reaction.…”
