Modulated TRPC1 Expression Predicts Sensitivity of Breast Cancer to Doxorubicin and Magnetic Field Therapy: Segue Towards a Precision Medicine Approach

Abstract: Chemotherapy is the mainstream treatment modality for invasive breast cancer. Unfortunately, chemotherapy-associated adverse events can result in early termination of treatment. Paradoxical effects of chemotherapy are also sometimes observed, whereby prolonged exposure to high doses of chemotherapeutic agents results in malignant states resistant to chemotherapy. In this study, potential synergism between doxorubicin (DOX) and pulsed electromagnetic field (PEMF) therapy was investigated in: 1) MCF-7 and MDA-MB… Show more

“…The employed magnetic paradigm produces a low level of oxidative stress that, while adaptive in nature in healthy tissues [ 29 , 64 ], may reach damaging levels in metabolically inflamed tissues [ 65 ]. Therefore, the exposure of an operated limb with aggravated and surgically-inflamed lesions may not have been the best therapeutic approach.…”

“…On the other hand, if taken into consideration, exposure regimes can be potentially designed to target a specific objective. For instance, we have recently shown using the same magnetic technology that stronger magnetic stimulation can be used to specifically halt breast cancer growth, employing the same molecular machinery [ 72 ]. Therefore, PEMF-based therapeutic strategies can be ultimately designed to either promote or arrest development depending on exposure intensity (duration, amplitude, and frequency) and the inflammatory status of the tissue in question.…”

“…The culturing of MCF-7 (HTB-22™) was previously described [ 38 ]. MDA-MB-231 cells were obtained from Dr. Glenn Kunnath Bonney, NUS, and maintained in Dulbecco’s Modified Eagle Medium (DMEM) (Hyclone; Danaher, Washington, DC, USA) with 10% FBS.…”
