Modulation of human cytochrome P450 3A4 (CYP3A4) and P-glycoprotein (P-gp) in Caco-2 cell monolayers by selected commercial-source milk thistle and goldenseal productsThis article is one of a selection of papers published in this special issue (part 1 of 2) on the Safety and Efficacy of Natural Health Products.

Abstract: In this study, we used an in vitro Caco-2 cell monolayer model to evaluate aqueous extracts of commercial-source goldenseal (Hydrastis canadensis) and milk thistle (Silybum marianum) capsule formulations, their marker phytochemicals (berberine and silibinin, respectively), as well as dillapiol, vinblastine, and the HIV protease inhibitor saquinavir for their ability to modulate CYP3A4 and ABCB1 expression after short-term exposure (48 h). Both upregulation and downregulation of CYP3A4 expression was observed w… Show more

“…Pellucidin A from P. pellucida was reported to have antimutagenic and antifungal effects against Trichophyton mentagrophytes [34]. Dillapiole has numerous reported effects; it has been found to act as a cytochrome P450 3A4 inhibitor, antibacterial agent, insecticide, antifungal agent, and anti-inflammatory agent in addition to DNA adduction activity [51][52][53][54][55][56][57]. Dillapiole that was isolated from P. pellucida was reported to have a gastroprotective effect [58].…”

“…As for moderation of CYPs on a transcriptional level, downregulation of CYP 3A4 was only reported in Caco-2 monolayer [47], but not in primary human hepatocytes [48].…”

“…Herbal interactions involving the CYP3A4 enzyme and P-gp may be responsible for many cases of therapeutic failures or adverse effects already reported, especially in patients under treatment with narrow therapeutic index drugs 11 , 18 , 19 . The impact of V. officinalis and P. methysticum on the activities of CYP3A4 and P-gp is a topic of debate among different authors, with some demonstrating inhibition 20 , 21 and others no effect 22 , 23 , 24 .…”
